J2抗双链RNA IgG2a单抗是双链RNA检测的金标准。起初只用于植物病毒检测研究,但从2006年Weber et al,发表文章表明:在检测的感染细胞内所有的正链RNA病毒均产生大量双链RNA,随后该抗体被应用于广泛的检测系统,发表文章超过200余篇。
J2抗体用于检测病毒dsRNA 中间体,例如:丙型肝炎病毒、登革热病毒、小鼠脑脊髓炎病毒、鼻病毒、基孔肯雅病毒、狂犬病病毒、脊髓灰质炎病毒、猪瘟病毒、雀麦草花叶病毒检测样本可以是细胞培养物或石蜡包埋的组织。
J2抗体被广泛应用于电镜学、免疫荧光显微技术、免疫组化及各种免疫捕捉实验,如:斑点免疫印迹、ELISA。J2抗体是用于区分被检样本是病毒或细菌的有效工具((Richardson et al., 2010))。近,J2用于制备mRNA过程中dsRNA的监测,因此该抗体基因治疗中,有潜在的应用价值。

K1单克隆抗体,IgG2a亚型,能够识别dsRNA(双链RNA)。K1抗体可用于检测细胞和组织中的dsRNA.经证明,K1抗体是J2很好的替代品。在一些特殊的实验中,用K1代替J2抗体,可以明显解决交叉反应或者是高背景。
K1抗体可以检测病毒dsRNA中间体,例如肝炎病毒、小鼠脑脊髓病毒、乙型脑炎病毒等。K1抗体还可以检测培养细胞以及石蜡包埋组织中的dsRNA。
如果要检测Poly I:C(多聚肌苷酸),我们强烈推荐使用K1抗体而非J2抗体,因为K1与合成的多核糖核苷酸有很强的亲和力。
K1抗体被广泛应用于免疫荧光、流式细胞术(FACS)和免疫捕捉检测,例如斑点免疫印迹及ELISA实验。

K2单克隆抗体,lgM亚型,是抗dsRNA的单克隆抗体。
K2抗体主要用于一些特殊的检测。K2抗体主要用于ELISA和夹心法ELISA实验,一般来说,K2抗体只能用于检测IgG(IgG2a)亚型抗体不适合的双链RNA(dsRNA)。
K2抗体需干冰运输,因为高浓度的K2抗体容易发生凝聚,因为不容易做成冻干粉,不能常温运输。(J2和K1可常温运输。)

温馨提示:
1)J2、K1单克隆抗体由于冻干过程中,稀释的抗体可能含有少量的变性蛋白聚合物,这些蛋白可能会对免疫组化实验产生高背景。因此,我们推荐在使用前,抗体重悬后,需离心,使用其上清。
2)J2、K1可常温运输,稀释后请分装,-20 ℃或-70℃保存,尽量避免反复冻融。
3)K2需干冰运输,到货后请分装,-70℃保存。
